In a blow ahead of the first Ashes Test, the 35-year-old made the decision after a chronic knee injury flared during the warm-up game against Essex.
Making his Test debut in 2010, Harris quickly became a lynchpin of the Australian attack.
He leaves the game with a record of 113 wickets in 27 Tests and 44 wickets in One Day Internationals.
