Heavily armed police have swooped on Melbourne nightclub Two Floors Up, arresting two men over an alleged theft.

Police, including members from the Critical Incident Response Team, swarmed the Lonsdale Street nightclub at 7am on Monday and took the two men into custody.

A large police presence remained around the venue until 10am, with officers blocking the entrance to the club and numerous police cars and vans parked nearby.

It's not the first time Two Floors Up, which has a 7am liquor licence and claims to be a "cornerstone in Melbourne's night clubbing scene", has made headlines.

Police allegedly uncovered thousands of ecstasy pills, methamphetamine and cash during raids on it and another dance club in April that led to serious trafficking charges.

The two men arrested on Monday, a 33-year-old from Glenroy and a 25-year-old from Broadmeadows, are being questioned in custody.
