Thieves have used a hammer to smash their way into an office of the nation's oldest animal welfare organisation to steal a charity box.

Police believe up to three offenders broke through a glass entrance at the RSPCA's Burwood East building on Thursday morning at 4.15am.

RSPCA Victoria chief executive Liz Walker said thieves took about $500 in coins and only had access to the reception area, and no animals or people were injured.
