A reclusive elderly couple with an interest in health and nursing has donated $35 million towards the University of Sydney's proposed health hub.

Property magnates turned philanthropists Susan and Isaac Wakil are behind the largest gift ever received in the University's 165-year history.

"We were inspired by the radical and innovative approach the University of Sydney is taking to address immediate and future healthcare challenges," Mr Wakil said in a statement on Thursday.

"Susan and I are pleased to be able to make this project a reality."

The Susan Wakil Health Building will house multiple health disciplines and provide clinical simulation programs and a multi-service clinic.

"Susan and Isaac Wakil's incredible generosity will harness greater collaboration between all the health faculties to deliver better healthcare to millions of people," Chancellor Belinda Hutchinson said.
