Police believe they know who is responsible for the shooting death of a 37-year-old man in Sydney's west.

They say the late-night shooting on Wednesday in Guildford is not linked to the city's recent spate of gangland killings and have formed Strike Force Buttle to investigate the murder.

Emergency services responded to reports of a shooting at 11pm at a house on Kennedy Street and found the wounded victim, who was reportedly shot in the chest.

Paramedics provided first aid, but the man died at the scene, police say.

Two or three people are believed to have barged into the home and shot the man in a targeted attack over a drug dispute, Fairfax Media reports.

Witnesses have told police they saw at least two people running from the scene.

The body was removed on Thursday morning and a crime scene remained in place during the day as officers interviewed neighbours.
