A man and a woman have been charged with murder over the fatal shooting of a man on an inner-Melbourne street on Tuesday afternoon.

A 43-year-old Noble Park man and a 21-year-old Yarraville woman were arrested in Dallimore Court, Noble Park, in Melbourne's south-east, about 5.30pm yesterday.

Their arrests come as part of police's investigation into the killing of 55-year-old Fitzroy man Brian Laidlaw on Napier Street in Fitzroy about 4.15pm on Tuesday.

Police believed the victim had been involved in an altercation with people in a black Audi parked on the street before he was fatally shot.

Detective Superintendent Paul O'Halloran told 3AW radio the victim was allegedly sitting in the car with two people he knew, before he got out and attempted to run away.

One of the people in the car then got out and allegedly shot him.

"The man was shot in the back and in the upper body," O'Halloran said.

"It was targeted and we believe he was known to (the gunman)."
