A black suitcase left on a bench outside Batman’s Hill Hotel on Collins Street is believed to be the focus of the police operation.
Authorities have advised motorists to seek an alternate route. Pedestrians have also been stopped from entering the street.
Trams have ceased operation along the street between Southern Cross Station and Batman Park. Routes 12, 96 and 109 are affected.
It is not known when the road will re-open.
