A five-year-old boy has died and a 12-year-old girl is fighting for her life after a single-vehicle crash in Victoria's east.

Four children and a woman were in the car when it left the road and rolled at Lindenow South just before 5pm on Sunday.

Police say a five-year-old boy has died and a 12-year-old girl is being flown to the Royal Children's Hospital with life-threatening head and chest injuries.

A 12-year-old boy will also be flown to hospital in a serious condition, while a third boy with minor injuries has been taken to nearby Bairnsdale for assessment.

The driver, a 32-year-old Lindenow South woman, was not injured and is helping police.

Police believe she lost control on a bend.

Victoria's provisional 2015 road toll stands at 116, up from 110 at the same time last year.
